Cellometer® Automatic Cell Counters - Auto T4 DataTotal Cell Count: Comparison to Hemacytometer Total Cell Count: Comparison to HemacytometerThe following figure depicts a typical comparison study between manual hemacytometer and Cellometer® Auto T4. The data indicates that same cell concentrations result from these two measurement techniques. SKBr-3 cells were suspended in media. For hemacytometer measurements, cells in all four-corner squares were counted. The following formula was used to determine concentration.
Cell viability measurement Auto T4 versus hemacytometerThree samples of mouse B cells with different viabilities were measured using hemacytometer and Cellometer® Auto T4. The viability ranged from approximately 27% to 87%. The Auto T4 measured viability was plotted against hemacytometer measurement. The resulting graph is shown as following: ![]() The results indicate that viability data measured by both methods are the same over a wide range of cell conditions. PBMC dilution seriesProcedure for Counting PMBC:
![]() Auto T4 images of fresh human PBMC cells with a background of a large number of platelets. In this case, PBMC cells are identified by size and counted, while platelets are excluded. ![]() Above figure shows data of PBMC dilution measurement. A total of 5 two-fold dilutions are used to generate the plot. Good correlation was obtained for dilution factor and measurement results. L929 cell dilution series for cell concentration and viabilitySample of L929 mouse fibroblast cells were diluted using trypan blue dispersed with growth media. Cell concentration and viability were measured using Cellometer® Auto T4. Both cell concentrations and viabilities were plotted against the dilution factors. With more dilution, cell concentration correlated to the dilution factors, while the viability remained the same.
