Transcript: Primary Cell Concentration & Viability using Cellometer Auto 2000 Cell Viability Counter

The Cellometer Auto 2000 Cell Viability Counter is specifically designed for accurate analysis of primary cells.

The user-friendly touch screen features pre-optimized assays for analysis of PBMCs, nucleated cells, splenocytes, cell lines, tumor cell suspensions, stem cells, and other primary cells. Users can also determine GFP transfection efficiency and viability.

Viability analysis can be performed with trypan blue or fluorescent dyes. In this example, acridine orange and propidium iodide are used to stain PBMCs. 20µl of stained sample is loaded into a Cellometer Imaging Chamber. The chamber is inserted into the Cellometer Auto 2000 and the desired assay is selected.

A dual-fluorescence assay is recommended for analysis of primary cells from heterogeneous samples, such as whole blood, cord blood, and bone marrow.

After previewing the image, a click of a button starts counting.

Acridine orange is a nuclear staining dye that stains all live nucleated cells, generating green fluorescence that is easily distinguishable from debris and unwanted cell types.

Propidium iodide enters dead cells with compromised membranes, generating red fluorescence.

In just 30 seconds, the cell count, concentration, mean diameter, and % viability are displayed on the screen.

A cell size histogram can also be viewed.

Brightfield images can be viewed to show cell morphology.

A fluorescent image showing green fluorescent, live counted cells and red fluorescent, dead counted cells can also be viewed.

Cell images and results can be auto-saved to a specified network location.

The all-plastic, self-contained counting chamber is easily removed after use, eliminating wash steps and potential contamination.
