Tips for producing MCF7 single cells
- Wash cells first with calcium free medium or a few ml of trypsin beforetrypsinizing
- Use trypsin with EDTA for trypsinizing which helps to chelate any calcium that isnecessary for cell adhesion
- Wait for about 5 or more minutes with trypsin
- Thoroughly pipette the cells up and down when adding medium following thetrypsin step, using 10 ml sterile pipette
Tips for counting very clumpy suspension cells
- Add 100uL of trypsin to 1mL of cell sample
- Mix up and down 10X
- Incubate 5-10 minutes
- Remove sample to count
